The MTT assay is defined as a method that measures cellular metabolic activity by assessing the reduction of MTT to purple formazan crystals primarily by mitochondrial dehydrogenases, providing insights into cytotoxicity levels in cells.

The MTT assay for cellular metabolic activity is almost ubiquitous to studies of cell toxicity; however, it is commonly applied and interpreted erroneously. We investigated the applicability and limitations of the MTT assay in representing treatment toxicity, cell viability, and metabolic activity.

MTT is used to assess cell viability as a function of redox potential. Actively respiring cells convert the water-soluble MTT to an insoluble purple formazan. The formazan is then solubilized and its concentration determined by optical density.
